A small cut with a big impact

Thursday, May 3, 2012

Diseases and injuries trigger warning signals in our cells. As a result, genes are expressed and proteins produced, modified or degraded to adapt to the external danger and to protect the organism. In order to be able to produce a particular protein, the corresponding DNA segment, the gene, needs to be expressed and translated. The DNA is localized in the cell nucleus, and exists as a long string that is coiled and bound by proteins. ARTD1 is one such protein, and therefore has the potential to regulate the expression level of genes through its interaction with DNA.

If cells detect warning signals or foreign bodies like bacteria and viruses in their surroundings, the expression profile of genes changes and an inflammatory response is triggered. To induce changes in gene expression, ARTD1 is removed from particular sites of the DNA. The process by which this is brought about has, until now, remained elusive. The team headed by Professor Michael O. Hottiger from the Institute of Veterinary Biochemistry and Molecular Biology at the University of Zurich has now discovered how ARTD1's DNA recruitment is regulated during inflammation, thereby influencing gene expression and subsequently inflammation.

Molecular scissors

As the researchers demonstrate in Molecular Cell, ARTD1 is cut into two pieces by molecular scissors, the protein caspase 7. Upon cleavage, these pieces can no longer bind to the DNA, thus allowing for more efficient gene expression.

The cleavage of proteins by caspase 7 was so far mainly associated with cell death. "The cleavage of ARTD1 by caspase 7 during inflammation constitutes a new biological function. It permits a new understanding of inflammatory responses and, in the longer term, may lead to the development of new anti-inflammatory drugs," explains Professor Hottiger. The results are of considerable importance because inflammation underlies most diseases, including cancer, immune disorders or metabolic syndrome.

Plant diversity is key to maintaining productive vegetation

ScienceDaily (May 3, 2012) ? Vegetation, such as a patch of prairie or a forest stand, is more productive in the long run when more plant species are present, results of a new study show.

The long-term study of plant biodiversity found that each species plays a role in maintaining a productive ecosystem, especially when a long time horizon is considered.

The research found that every additional species in a plot contributed to a gradual increase in both soil fertility and biomass production over a 14-year period.

This week's issue of the journal Science published the results. They highlight the importance of managing for diversity in prairies, forests and crops, according to Peter Reich, lead author of the paper and a forest ecologist at the University of Minnesota.

Reich and colleagues looked at how the effect of diversity on productivity of plants changed over the long-term.

Two large field experiments were conducted at the National Science Foundation's (NSF) Cedar Creek Long-Term Ecological Research (LTER) site in Minnesota, one of 26 such NSF LTER sites around the globe in ecosystems from forests to grasslands, tundra to coral reefs.

"This study reveals what short-term experiments have missed: that the effects of biodiversity loss on ecosystems are more complex, severe and unpredictable than previously thought," says Matt Kane, an NSF LTER program director.

"The work shows the importance of doing long-term research," says Kane, "in this case documenting for the first time the critical importance of biodiversity for ecosystem health and sustainability."

The biodiversity experiments at Cedar Creek are the longest-running such experiments in the world, says Reich.

They contain plots with one, four, nine or 16 different species of plants.

The research used long-lived prairie plants, but serves as a model system for all vegetation, whether prairie, forest or row crop.

The study also showed how diversity works by demonstrating that different species have different ways of acquiring water, nutrients and carbon--and maintaining them in an ecosystem.

"Prior shorter-term studies, most about two years long, found that diversity increased productivity, but that having more than six or eight species in a plot gave no additional benefit," Reich says.

The scientists found that over a 14-year time span, all 16 species in the most diverse plots contributed more and more each year to higher soil fertility and biomass production.

"The take-home message," says Reich, "is that when we reduce diversity in the landscape--think of a cornfield or a pine plantation or a suburban lawn--we are failing to capitalize on the valuable natural services that biodiversity provides."

Co-authors of the paper are David Tilman, Forest Isbell, Kevin Mueller, Sarah Hobbie and Nico Eisenhauer of the University of Minnesota, and Dan Flynn of the University of Zurich.

Romney looks to Virginia governor for assist in swing state

PORTSMOUTH, Virginia (Reuters) - For Republican Mitt Romney, it was another visit to a battleground state, another appearance with a potential running mate - and another endorsement from a former rival who once had seethed at the idea of Romney being elected U.S. president.

Former Massachusetts governor Romney stopped in this southeastern Virginia city on Thursday and looked to Republican Governor Bob McDonnell for help in inspiring voters in the politically divided state, which Romney hopes to reclaim for his party in the November 6 presidential election.

Romney's visit to Virginia - one of 10 "swing" states that are likely to decide this year's presidential election - was a reminder of the potential and pitfalls the presumed Republican nominee faces in Virginia, which Democratic President Barack Obama won en route to winning the 2008 election.

Polls indicate Obama leads Romney in the state, where McDonnell's popularity among voters put him on many short lists of potential running mates for Romney.

But then came the bruising Republican presidential primaries, when Romney was reluctantly drawn into discussions of divisive social issues such as abortion and contraception.

McDonnell became part of the story by promoting state legislation that would have required women to undergo an invasive transvaginal sonogram before getting an abortion.

The bill led to protests at the state Capitol in Richmond. A revised version McDonnell signed into law requires ultrasounds prior to abortions, but less invasive abdominal ultrasounds are an option for Virginia patients.

It's unclear whether the controversy affected McDonnell's chances of becoming Romney's running mate, but on Thursday the Virginia governor was an enthusiastic soldier for Romney.

While introducing Romney, McDonnell reminded a crowd of Romney supporters about Obama's 2008 campaign theme of bringing hope and change to the country.

"Remember three and a half years ago, we heard that tune about 'hope and change?' And now what do we have? We have recession, division and malaise," McDonnell said. "It's time for a change, don't you think?"

BACHMANN REVERSES COURSE

McDonnell later told Fox News he would consider being Romney's running mate if asked, but that he was not interviewing or auditioning for the role.

"I'd say that's good media drama, but that's not what has happened," McDonnell said. "I just believe we need a change in leadership in this country with our job picture, our debt picture."

Also standing with Romney was U.S. Representative Michele Bachmann of Minnesota, a former rival of Romney in the Republican primaries.

Earlier in the Republican primary , Bachmann had said Romney "cannot beat Obama" because the president's healthcare overhaul - a target of conservatives - was based on a healthcare program Romney imposed in Massachusetts.

On Thursday, Bachmann said Republicans should support Romney because he could help jumpstart U.S. energy production, cut high gasoline prices and create jobs.

"Mitt Romney's future for America would be a legalization of American energy," she said. "It's something to get excited about. It's why we must elect Mitt Romney as the next president of the United States."

Her endorsement came a day after that of former U.S. House Speaker Newt Gingrich, who had been a bitter rival of Romney in the Republican primaries.

Romney - who in recent days has toured several swing states with potential running mates including Florida Senator Marco Rubio, New Hampshire Senator Kelly Ayotte, Ohio Senator Rob Portman and Wisconsin Rep. Paul Ryan - continued to focus on economic issues, the core of his bid to defeat Obama on November 6.

In Portsmouth, Romney said Obama's halting of oil drilling off the Virginia coast had hurt job creation in what he said was a state that could decide "who the next president is."

"Right here in Virginia, the idea that you have powerful energy right off your coast, that you could be creating good jobs, that's been lost by a president who says no," he said.

Obama opened up offshore oil drilling in Virginia in March 2010 in an effort to gain support for his clean energy strategy. But the president reversed course two months later, after the massive BP oil spill in the Gulf of Mexico.

CONTROVERSIAL RESIGNATION

After several presidential elections in which Virginia backed Republicans, Obama won the state in 2008, reflecting a trend toward Democratic candidates driven largely by growth in the northern Virginia suburbs near Washington.

Also Thursday, Romney's campaign continued to come under criticism for its handling of a situation involving Richard Grenell, an openly gay man who resigned this week as the campaign's foreign policy spokesman after anti-gay conservatives complained about his hiring.

On Tuesday, Romney campaign manager Matt Rhoades said the campaign was "disappointed" at the departure of Grenell, a former U.S. spokesman at the United Nations. "We wanted him to stay because he had superior qualifications," Rhoades said.

The New York Times reported Wednesday that top Romney campaign staff members tried to persuade Grenell not to resign but that he was frustrated after being asked by the campaign to keep a low profile until the public relations flap died down.

Before joining the Romney campaign, Grenell was a prolific and inflammatory voice on Twitter. But once he got the job he deleted his most provocative musings on Twitter, which had included messages aimed not just at Democrats and the media but fellow Republicans.

Some questioned whether the Romney campaign, known for being meticulous in its planning, had failed to properly vet Grenell.

But some gay rights activists said the real issue was the campaign's hesitancy to publicly support Grenell when he was being criticized by social conservatives.

"There are choices to be made in campaigns," said Jimmy LaSalvia, executive director of GOProud, a gay Republican group in Washington. "They chose to be quiet in hopes that it would blow over, and it festered and got worse. Had they confronted it, it wouldn't have become the issue that it did."

(AP) ? A teenager blew himself up near a Pakistani market close to the Afghan border Friday, killing 20 people, officials said. The suicide bombing came a day after the U.S. released letters seized from Osama bin Laden's compound that criticized Pakistani militants for killing too many civilians.

Five of the dead in the blast in the northwestern Bajur tribal area were local members of the security forces, including one who had received an award for bravery in fighting Islamist militants, government administrator Abdul Haseeb said. The others were passers-by, including a woman and several schoolchildren.

The suicide bomber, who was believed to be 15 to 16 years old, detonated his explosives as he approached a security checkpoint near the market in the town of Khar, said Haseeb. The attack took place as locals were headed to work or taking their children to school. More than 40 people were wounded, most civilians.

The explosion damaged more than a dozen shops, and rescue officials searched through the debris for anyone trapped inside. The ground outside the market was covered in blood and littered with the shoes of those hit by the attack.

Pakistani Taliban spokesman Ahsanullah Ahsan claimed responsibility for the bombing in a telephone call to The Associated Press.

"This is revenge for killing our people," said Ahsan.

Pakistan's branch of the Taliban, which is battling for control of the region, has killed and wounded thousands of people since 2009, many of them civilians in markets or mosques.

On Thursday, the U.S. released 17 letters seized from Osama bin Laden's compound by American commandos after they had killed the al-Qaida leader a year ago. Many of them addressed bombings in which civilians were killed.

Two senior al-Qaida leaders wrote a letter to Pakistani Taliban chief Hakimullah Mehsud in December 2010 criticizing the group for carrying out attacks in markets, mosques and other places that kill innocent Muslims. It accused the group of making "clear legal and religious mistakes that might result in a negative deviation from the set path of the jihadists' movement in Pakistan."

The group's actions also "are contrary to the objectives of jihad (holy war) and to the efforts exerted by us," said the letter, which was written by Abu Yahya al-Libi and a militant leader identified as Atiyatullah.

A bin Laden spokesman, the American Adam Gadahn, wrote a letter to an unknown recipient in January 2011 in which he listed a series of attacks on mosques and other sites in Pakistan that killed hundreds of civilians. The Pakistani Taliban either claimed responsibility for the attacks or were suspected of carrying them out.

Gadahn said these kinds of attacks "distort the picture of the pious and loyal mujahedeen."

"Now many regular people are looking at the mujahedeen as a group that does not hesitate to take people's money by falsehood, blowing up mosques, spilling the blood of dozens of people in the way to kill one or two who were labeled as enemies," said Gadahn.

One of the incidents highlighted by Gadahn was a 2010 suicide bombing of a mosque in the northwest town of Darra Adam Khel targeting anti-Taliban tribal elders that killed nearly 70 people. He expressed skepticism at attempts by the Pakistani Taliban to blame the attack on the private U.S. security firm formerly known as Blackwater.

The Pakistani Taliban rarely claim responsibility for attacks that kill many civilians, often blaming them on the U.S. or Pakistani governments.

Gadahn advised the Pakistani Taliban to carry out more targeted attacks that would limit civilian casualties.

"If that targeted person really deserves to be killed, why not employ another method rather than random attacks, which are not tolerated by any conscience or religion?" said Gadahn. "Have you not remembered that you are fighting in the Muslim towns and not in the infidel's fortresses?"
